Plain-language answer
Gangliosides are a special group of glycosphingolipids whose sugar chains carry one or more units of sialic acid, an acidic sugar that gives them a negative charge. They are especially plentiful in the brain and nerves, where they make up a large share of the sugar coating on nerve cell surfaces.1
Because gangliosides are so concentrated in the nervous system, they matter for how nerve cells develop, connect, and stay healthy. Their surface sugars are recognized by the immune system, and in some nerve conditions the body mistakenly makes antibodies against its own gangliosides. They also serve as landing sites that certain bacterial toxins use to enter cells.1
Technical detail
Gangliosides are sialylated glycosphingolipids of the ganglio series, most abundant in neural membranes, synthesized by sequential sialyltransferase and glycosyltransferase action in the Golgi, and named under the Svennerholm system by sialic acid count and migration.1
Gangliosides are built on a lactosylceramide core extended into the ganglio series and decorated with one or more sialic acid (N-acetylneuraminic acid) residues. Under the Svennerholm nomenclature the letter records the number of sialic acids, so GM, GD, and GT denote mono-, di-, and tri-sialylated species, and the numeral and subscript indicate the core length and isomer, as in GM1, GD1a, and GT1b.1
Gangliosides are enriched in the plasma membranes of neurons, where their clustered, negatively charged glycans contribute to membrane microdomain organization, modulate cell adhesion and receptor signaling, and are engaged by glycan-binding proteins including the sialic-acid-recognizing Siglecs. Their prominence on nerve surfaces makes them targets of both microbial toxins and autoimmune antibodies.1
Human relevance
In Guillain-Barre syndrome and related neuropathies the immune system produces antibodies against nerve gangliosides, and molecular mimicry between microbial glycans and ganglioside structures is a recognized trigger of this anti-ganglioside response.1
